PHILADELPHIA Dual inhibition of vascular endothelial growth factor and c-MET signaling inhibited tumor invasion and metastasis in a laboratory model of pancreatic neuroendocrine cancer, according to a paper published in Cancer Discovery, the newest journal of the American Association for Cancer Research.
"Inhibition of VEGF signaling plus c-MET signaling results in a synergistic effect on tumors that leads to slowing of tumor growth and decreased invasiveness and metastasis," said lead researcher Donald M. McDonald, M.D., Ph.D., a professor at the University of California San Francisco Comprehensive Cancer Center.
Previous laboratory research had shown that inhibition of VEGF signaling with agents like bevacizumab or sunitinib can give rise to a number of side effects including increased tumor invasion and metastasis.
What was not known was whether anti-VEGF therapy results in elevated c-MET expression, which has been previously shown to promote tumor cell invasiveness and metastasis. To determine this, McDonald and colleagues conducted a two-phase laboratory study. They treated mice engineered to develop pancreatic neuroendocrine tumors with an anti-VEGF antibody, which reduced tumor size but increased invasiveness and metastasis. This treatment also increased tumor hypoxia and expression and activity of c-MET.
However, when both VEGF and c-MET signaling were inhibited simultaneously, there was a reduction in invasion and metastasis. The researchers tested three c-MET inhibitors: crizotinib and PF-04217903, which target c-MET but not VEGF signaling, and cabozantinib, a dual inhibitor that blocks VEGF and c-MET signaling.
McDonald said they conducted their initial study in neuroendocrine pancreatic tumors because the genetic mouse model of these tumors has been studied so extensively, and then observed the effect in other tumors as well.
"The intent of this study was to explore a mechanism, and there is no indication that this effect will be confined to pancreatic tumors," he said.

North Greenville offers a variety of intramural sports, including Flag Football, Co-ed Sand Volleyball, Co-ed Ultimate Frisbee, Dodgeball, Disc Golf, Spades Tournaments, Men?s Softball, Women?s League Basketball, Competitive League Basketball, Recreation League Basketball, Co-ed Softball, Competitive League Indoor Soccer, and Recreation League Indoor Soccer.
The goals of NGU intramural sports as described on the school website are ?to place an emphasis on sportsmanship, teamwork, and personal fitness within the context of a Christ-centered environment. Everyone is invited to participate, regardless of skill level or past experience. Intramural sports are fun, free, and an excellent way to build relationships with those possessing similar interests.?

It's important to parse the distinction between the 9th Circuit striking down the unconstitutionality of overturning existing state law and them saying that opposing gay marriage is in itself unconstitutional (which they did not).
Still the 9th Circuit Court's ruling for not the first time revealed the pettiness and lawlessness behind the homophobic mindset that created Prop H8 and has kept this useless referendum on the books for over three years. Said Judge Stephen Reinhardt in his majority opinion,
Proposition 8 serves no purpose, and has no effect, other than to lessen the status and human dignity of gays and lesbians in California, and to officially reclassify their relationships and families as inferior to those of opposite-sex couples.
As I said last night in my jeremiad to GOProud, the hateful organization that discriminates against liberals as everyone but liberals discriminates against them, the only thing distinguishing the LGBT community from everyone else is with whom we choose to sleep. And with whom we fall in love. Falling in love, as virtually any human being will tell you, is as involuntary as our sexual identities.
It's the love part that freaks out right wingers and homophobes so much they cannot even bring themselves to address it. This is why they confine their arguments to gay sex, something guaranteed to give their fellow homophobes the dry heaves. They faithfully trot out Leviticus, a book in the Bible that was plainly written by deeply disturbed people, as if it's the law we ought to be observing and enforcing including the capital punishment part.
It's the narrowing of their argument to anal sex, swallowing semen, one woman going down on another woman and AIDS that produces a visceral reaction in others who are simply incapable of believing that sexual orientation is something you're born with. Love? Well, that's much, much harder to invalidate. We may not have experience in gay sex and many of you may even feel it's repugnant. But most of us, if we're lucky, know what true love feels like.
So, automatically defeated on that front, they narrow their arguments as they seek to criminalize homosexuality, even going as far as to import their hateful homophobia to other countries with the express purpose of imposing the death penalty.
And as if discriminating against their fellow humans isn't bad enough, the GOP is taking more than baby steps toward genocide by decriminalizing anti-gay bullying and even physical violence. When the Michigan Senate voted on an anti-bullying bill, House Republicans had already succeeded in inserting an amendment that neatly defanged the legislation and even subverted its entire reason for being. Michigan Republicans made it legal for one student to bully, beat and even kill a gay, lesbian or bisexual student as long as they whip out the Religion/Morality card.
Essentially, it turned Matt's Safe School Law into a 007 license for homophobes to beat gay students with absolute impunity. Even more despicably, this was hailed as a "compromise" with Republicans, despite it offering no real protections for LBGT youth who since time immemorial have been specially singled out for persecution.
This genocidal movement against LGBT youth is multi-faceted. Right wingers like Tony Perkins of the Family Research Council screamed until they were blue in the face over President Obama making Kevin Jennings the head of the Office of Safe and Drug-Free Schools. Jennings achieved notoriety in right wing circles by founding the Gay, Lesbian and Straight Education Network, which caters to straight students as well as LGBT youth. Mr. Jennings' responsibilities as President Obama's Safe Schools Czar also includes curbing drug abuse.
When you see and hear the furor arising from Mr. Jennings' appointment by our president (think about that, too, GOProud, next time you try to spit venom at liberals and Democrats who allegedly hate gays), you shouldn't need to take even half a step back in the interests of perspective, objectivity and context to see how hateful it is for anyone to stand in the way of making our public schools safer for our greatest asset and resource: Our youth. Our very future.
Religious freedom, to people like Perkins and other evangelical homophobes, essentially boils down to getting their way every time and not being disagreed with even at the expense of endangering our LGBT youth and indemnifying their abusers and killers.
If they're willing to disrespect our nation's LGBT youth, then no one with an ounce of compassion and grey matter should feel beholden to respect their homophobia whether or not it's clutching a cross. The First Amendment curtails the government's attempts to establish a national religion and mandates that it respect all faiths. It does not obligate individuals to do the same.
I do not respect a religion that is not all-inclusive and seeks to destroy a portion of humankind that has been around since the earliest days of prehistory and will be around as long as our species endures. And while my passion for certain issues waxes and wanes, what will never diminish and what will the last thing for which I stop fighting, is gay rights for my people.
And if you evangelical nut jobs out there want to scream discrimination, I have but two words for you: "How does it feel?"

Merkel, who postponed a trip to Rome where she was to hold talks on the euro zone's debt crisis, voiced "deep regret" at his resignation. She moved quickly to limit the fallout and try to ensure a smooth succession, saying she would seek an agreement with the main opposition parties on the next president.
The chancellor is riding a wave of popularity in Germany for her handling of the crisis, but the departure of Wulff raises questions about her judgment because she forced through his appointment in 2010 over a strong opposition candidate most Germans favored.
Wulff is the second president to step down in less than two years. His predecessor, former International Monetary Fund chief Horst Koehler, resigned unexpectedly after coming under fire for comments he made about the German mission in Afghanistan and failing to get strong backing from Merkel.

Calling for Parliament to lift Wulff's immunity was an unprecedented move against a German president.

Since then, there has been a constant stream of new revelations that have chipped away at his credibility, leading the German media to mock him and even invent a new verb in his honor. "Wulffen" ? or literally "to Wulff" ? means to be evasive without telling a clear lie.
Wulff said in his resignation he was convinced he would be fully cleared of any wrongdoing.
"I have always behaved legally correctly in the offices I held," he said. "I have made mistakes, but I was always honest."
Wulff's longtime spokesman, Olaf Glaeseker ? whom the president fired in December ? is also under investigation on corruption allegations in connection with the organization of business conferences.
The resignation is likely to embolden the opposition Social Democrats (SPD) and Greens, who have shied away from criticizing Merkel too strongly in recent months.
Despite her vow to find a consensus candidate to replace Wulff, the choosing of a successor could prove divisive, distracting her just as European governments are trying to cobble together a second aid package for Greece to avert a chaotic default and euro zone exit.
Europe's economy edges toward recession
How much of a distraction it becomes depends on how fast a successor can be agreed upon. A vote in the 1,244-seat Federal Assembly must take place within a month, or by March 18.
"This won't be without consequences for Merkel, her reputation will suffer from it," said Gerd Langguth, political scientist at Bonn University.
Merkel won a second term in 2009 and will not face another federal election until the autumn of 2013. But her party faces battles to hold onto power in the states of Saarland and Schleswig-Holstein in regional votes due in March and May.
A member of Merkel's Christian Democratic Union (CDU) who served as premier of the western state of Lower Saxony, Wulff was once seen as a potential rival to Merkel and many in Germany saw his appointment as a ploy by the chancellor to push him out of the political arena.
Effigy
Wulff has been one of the main targets at this year's carnival celebrations in the Rhineland where politicians are traditionally lampooned.
One float in the city of Mainz has an effigy of him with a black eye and plaster on his forehead slumped at the edge of a boxing ring. In Cologne, where the biggest processions take place, Wulff is dressed up as a grey rabbit on the butcher's table about to be carved up.
On a trip to Rome earlier this week and in a briefing with a small group of journalists on Thursday, Wulff made clear he planned to hang onto his post.
The speaker of the upper house of Parliament ? Bavarian governor Horst Seehofer, a member of Merkel's conservative bloc ? now takes over the presidential duties on an interim basis, mostly signing legislation into law.
The leading contender to succeed him permanently is Joachim Gauck, an anti-Communist human rights activist in East Germany who ran against Wulff in 2010 and embarrassed Merkel by forcing the election in the Federal Assembly into a third round.
Other potential candidates include Defense Minister Thomas de Maiziere, Labour Minister Ursula von der Leyen, Bundestag President Norbert Lammert and possibly Finance Minister Wolfgang Schaeuble, though shifting him to Bellevue palace would leave a gaping hole in Merkel's cabinet in the midst of the euro zone sovereign debt crisis.
